WE ACCEPT SAMPLES AT ANY STAGE OF THE PIPELINE

Sample input recommendations services are available to maximize your project workflow and the results for your sequencing projects. Above all, these recommendations help maintain sample integrity, ensure traceability, and enable workflow efficiencies in the laboratory.

In effect, following these sample input recommendations will help ensure that the data generated from laboratory analyses is accurate, reliable, and reproducible. This is fundamental for advancing your scientific research and making informed decisions based on the results.

The following are SeqMatic’s recommendation for sample input for our library generation services. Please consult with your SeqMatic contact for assistance if your samples cannot meet our requirements.

Sample Cells or Tissues:
  • Please discuss with SeqMatic scientist regarding tissue quantity required
  • We accept samples in a variety of formats, please contact SeqMatic for more information
  • For RNA, samples can be flash frozen or stored in a RNA preservation solution such as RNA Later
Genomic or Amplicon DNA:
  • Recommended Volume: 20uL or more
  • Recommended Concentration: 20 ng/uL or higher
  • Minimum Volume: 10uL
  • Minimum Concentration: 10 ng/uL
  • A260:A280 value should be between 1.8 and 2.0
  • DNA should be buffered in 10mM Tris-HCl or TE buffer
  • Amplicon DNA should be free of PCR primers and dimers
Purified RNA:
  • Recommended Amount: 1000ng or more
  • Recommended Volume: 20uL or more
  • Recommended Concentration: 20 ng/uL or higher
  • Minimum Amount: 100ng (if less than 100ng RNA is available, discuss with SeqMatic scientist prior to sample submission)
  • Minimum Volume: 10uL
  • Minimum Concentration: 5 ng/uL (if less than 5 ng/uL, discuss with SeqMatic scientist prior to sample submission)
  • A260:A280 value should be between 1.8 and 2.0
  • RNA should be stored in nuclease free water or RNAstable.
  • RNA must be validated using an assay such as a BioAnalyzer RNA chip. RNA QC services are available for a fee.
Prepared Sequencing Library (MiSeq & NextSeq):
  • Recommended Volume: 20uL or more
  • Recommended Concentration: 10nM or higher
  • Minimum Volume: 10uL
  • Minimum Concentration: 2nM
  • Libraries should be pooled prior to submission. Library pooling services are available for an additional fee.
  • Provide at least 10uL of 100uM custom sequencing primers, if required
Prepared Sequencing Library (NovaSeq):
SP/S1 S2 S4
Recommended Volume: 200+ uL 250+ uL 350+ uL
Recommended Concentration: 10+ nM 10+ nM 10+ nM
Minimum Volume: 100uL 200uL 300uL
Minimum Concentration: 1nM 1nM 1nM
  • Libraries should be pooled prior to submission. Library pooling services are available for an additional fee.
  • Provide at least 20uL of 100uM custom sequencing primers, if required
Prepared Sequencing Library (NovaSeq Xp Lane Loading):
SP/S1 S2 S4
Recommended Volume: 50+ uL 50+ uL 60+ uL
Recommended Concentration: 10+ nM 10+ nM 10+ nM
Minimum Volume: 20uL 30uL 40uL
Minimum Concentration: 1nM 1nM 1nM
  • Libraries should be pooled prior to submission. Library pooling services are available for an additional fee.
  • Provide at least 20uL of 100uM custom sequencing primers, if required
